No, it won’t. As far as I’m aware there’s no provision for creating third-party applications for this device.

I just read your comment again. Do you mean to say that all of the apps that are on the WDTV are written by Western Digital (Hulu, netflix, MLB, ...)?
wow, that seems really locked down compared to all of their past boxes.
Perhaps I don’t know what I am talking about exactly, but doesn’t WD actually maintain an open-source alternative to their “Official” firmware on WDTV? (http://support.wdc.com/product/download.asp?groupid=1011&sid=161&lang=en) Even if not - I am sure I have previously seen several modified WDTV firmware packages that added features the official box didn’t have.
I personally own 3 of these boxes, and would be super-interested in seeing Plex service on them.

If there’s ever a supported SDK for writing third-party apps for the WDTV boxes, we’d definitely look into it! We’re not interested in creating apps that require that the device is “hacked” though, as the hardware manufacturer can dramatically change how things work between firmware updates, resulting in a constant cat & mouse game (and dissatisfied customers when the app stops working). See Apple vs. the jailbreakers for a good example 
There is a hacked firmware for the WD Live Plus called WDXLTV (google it). Basically it turns the WD LIVE into a linux box with a web interface… I dont know if you would be able to run plex on it, however, i dont see porting the linux release to it as being that big of an issue… I may look into this and let you guys know.

Any new updates on this? And for those of you who do own a WD Live, how is the experience with using Plex? I’m not a big fan of using the XBox or a first gen Roku and am considering this route.

I’ve posted this topic in the official wd tv play forums. People were nice enough to respond. It looks as though given that the wd is a closed platform there will have to be a collaboration between PLEX and WD or WD would have to open it up (to a least the PLEX guys).
So unless there is demand (a lot) I doubt this will happen
