So if you have both, then just plug it in and try it. Doing so can cause no harm.
But my guess is that it won't work, since it doesn't work with a PS2 gamepad adaptor I tried.
The RPi and RasPlex are designed to work with USB keyboards, and some other USB devices, but gamepads have an entirely different device profile, and I don't think RasPlex has the necessary device drivers to use them.

I tested it and nothing happens if I press a button on the pad. The controller also doesn't show up in the settings.

You'll need to write a new keymaps.xml, search the forums for examples of what to do, or Google, duckduckgo or whatever search engine you use.
