With the release of Nvidia RTX Video Super Resolution, I wonder how this works with Plex on desktops? I’d imagine the web client would benefit from this, whereas the native windows app wouldn’t. At least for now…

Probably somehow, but don´t expect it to be coming anytime soon. First, all layers above plex (e.g. ffmpeg) need to gain support for this and then plex needs to adopt it. Looking at the Plex FFmpeg update frequency, we could call this “very unlikely”.
I guess the easiest way we can get RTX VSR is with the Desktop players and a replaced mpv.dll when mpv gains support for RTX VSR. In the meantime, if you really really need it, a combination of Kodi with DirectShow support, the PlexForKodi Addon and the MPC-VR RTX SuperRes filter (as noted in the link above) might work. (although I don´t think it is reliable)
If you set the Video Quality to the highest level in the Plex Desktop and Plex HTPC apps, you already get some kind of advanced upscaling algorithm applied.
It’s the main reason for the high GPU load you experience with these apps.
