No issues here. Flashed the SD card and USB. Changed cmdline.txt file on the SD card with the I tried this "boot=/dev/sda1 disk=/dev/sda2 quiet oprofile.timer=1" and it booted fine.
I just did this with 0.3.0 and it worked fine.

This worked great for me and was able to play higher bitrate video without a problem. My limit before this was around 8Mbps and worked perfectly with a 10Mbps file. Might crank a sample up and see what it can take :-)
I did have a problem when I restarted it errored with a '...file already exists...' error and would not proceed any further. I did have it powered via the TV but now powered to always on. Can anyone else replicate this?

By label the USB drive and SD card, do you mean name them that when formatting the drives?
Yes that is what he means. I got It working with the new cmd line however SSH seems to not work even after adding SHH to the command line. Any got SSH working on 0.3.0 or higher with a USB.

Is browsing around in the menus and sections faster now when you run it from the USB?
i find it faster yes, also no more corrupt sd cards :)
I flashed the USB drive and the Pi is now booting from SD and running from USB. No noticeable improvements, seems a little slower overall if anything. Going to give it some time to cache and see how it is then.
Make sure the USB device is a speedy one, otherwise you gain nothing from this.

However, I'm going to try with a Transcend Jetflash 700 16gb (ordered and on its way) as it's only $15/GBP10ish - for reference, an 8gb was about $13 at the time of writing so I went one up.. Hope that works but worse comes to worse I just have to upgrade.

I tried using the windows installer on both my sd card and usb drive, but they have both been formatted at 249MB when they are both 16GB, is there any way to get the installer to format the whole card/drive?

On OpenELEC I used one of these http://www.amazon.co.uk/Lexar-JumpDrive-Flash-Drive-Memory/dp/B00B0YW9CW/ref=sr_1_1?ie=UTF8&qid=1382537610&sr=8-1&keywords=lexar+s23 pretty cheap but with really good performace, OpenELEC installed with one of those is a lot more responsive than I have managed to get RasPlex with just a class 10 SDHC, which is why I'm trying to get the USB working on RasPlex as well, see if it helps, but the installed formats the drive at just 249MB for both the sdhc and the usb drive I guess I'm going to have to do it manually as losing 15GB of space on both, so 30GB in total seems a waste.
249mb is the boot partition, if you look at drive with a program like GParted it will show you the other partitions
Rasplex works off of USB drive, where are you having problems getting this to work ?
Also no need to spend extra on USB3.0

there is a further 1.2gb partition on both, and 13.47 unallocated space on both
you can use GParted to expand the EXT4 partition

That is the one on USB correct, I added the SSH to it to reenable it, the sdcard has a different config
to clarify between config files -
SDcard - "boot=/dev/sda1 disk=/dev/sda2 ssh quiet oprofile.timer=1"
USB flash - "boot=/dev/mmcblk0p1 disk=/dev/mmcblk0p2 ssh quiet"
