As the title says, this combination results in no playback. Issue is also present on web player except the file will play for 1 second then stop (no audio).
Works well through desktop player and iOS apps.

The XBOX One does not support MKV files in it's current state. This will be added in the October update, but I am not sure if this will apply to third party apps such as Plex.
We will be able to take advantage of support that MS adds for other formats, so we're hoping it will be available in a future release.
PMS should be able to transcode your video file - although if you're having issues in web it suggests something is amiss. Please can you post some logs of when you attempt to play the video? https://support.plex.tv/hc/en-us/articles/200250417-Plex-Media-Server-Log-Files

It transcodes the DTS to 6 channel AAC and plays fine. Most of my MKVs don't have AAC audio as MKVs are usually DTS/AC3 and MP4s are 'usually' AAC.
Most MKVs are also playing just fine for me...with the exception of a few hiccups
Mine transcodes pretty much everything in .MKV format....because I'm running on a Synology NAS, it doesn't have the beef to actually transcode it quickly with little to no stuttering/impact. Because of this its not quite usable for me at the moment.... :(
Still a BIG fan of Plex, though, and am hopeful this can/will be resolved in the future! Go Plex team!

I'm running on a Drobo-5N ... I'm pretty sure the NAS is not your issue
For all of you who have trouble to play mkv files with xbox one app I had the same issues. Uninstall PMS completely and delete the settings folder, then reinstall PMS and it should work now.

Just to update, it looks like the complete removal of Plex and installing the most current one has helped quite a lot. There's still some stuttering issues on some of my higher quality content I have to sort out, but that's a far cry from "nothing works, help". :)
Once it finishes updating overnight, I will test and see if that was due to indexing, or some other limitation.
