Yes and No.
Yes the file will stream from the PC to the Xbox One. The Plex cannot switch the display mode so you would need to manually switch your TV into 3D mode during streaming.
There is also a weird behavior in the Xbox One media player that if sometimes detects the video stream as being a 3D stream it automatically switches playback mode into 2D so you cannot view the movie as 3D.
Other considerations:
Subtitles - You need to have subtitles designed for 3D playback. If you try and use automatically downloaded ones from Plex, they will not work properly.
Transcoding - If the video transcodes you may run into issues where the transcoded video doesn’t have a perfect split of the image which can mess with the 3D effect. (Normally, I have only seen this issue when the original video stream has a black strip separating the two images.)

Sorry but this wasn’t even true at the time you wrote it. Apps are allowed to switch display modes for things like refresh rate (24fps) and stereoscopic. See this article
But you guys know that because the last time you made this claim, someone who is a developer even told you how to do it.
Interestingly, that thread was locked. It says it was “automatically locked” because their were no replies in 90 days even though older threads with no replies (for example, this thread) weren’t locked. So can 3D switching be put onto the feature request list since the apps are allowed to do that now (and have for two years)? Do we have to publicly shame you guys the same way Netflix had to be in order to add 24fps support?
Sorry, but you are simply wrong. That article you linked to explicitly states :
“While frame rate switching works perfectly well for Blu-ray playback, Xbox One is locked at 60Hz for apps.”
The Plex app is an app which would mean it suffered from being locked at the 60Hz rate. This was the case for apps written on the original development framework for the Xbox One.
Further down the same author stated:
“As such, I contacted an UWP (Universal Windows Platform) developer, Pasquale Pignataro"
The Universal Windows Platform was not originally a part of the Xbox One development system and was released at a later date. This meant that apps there were written on the original framework were not able to take advantage of the newer API and features. This developer that the author spoke to worked on the new framework that did have these API’s. So instead of getting to the bottom of why older apps could not change the rate, he instead simply found out new apps could. To take advantage of the new API’s an app had to be rebased to use the UWP which is why the current version of the app replaced the older version of the app.
The very first sentence of that post starts with “If Plex is using the UWP API’s” which was not the case when I posted in this topic 2 years ago. That means the properties he mentioned were not available in the framework the app was using. So, yes I knew it was possible for newer apps to use the newer API’s, but the older version of the app could not and the method this person posted was unavailable for the older app to use.
Nothing interesting about it. Extremely old threads were migrated from an older forum manager so they weren’t ever posted to the current forum manager. That means threads like this (Which is over 2 years old) aren’t locked because they never had a chance to reach automatic locking period.
The reason I didn’t reply again in that post is because after I forwarded the information onto the developers, I moved on to new topics. And since no one else replied the topic locked.
I am not an employee of Plex, just a volunteer that tries to help out. As far as I know they are aware of the desire for 3D playback automatic switching, but I do not have access to their development road map so I cannot see what has been added or what is currently being worked on.
