I created an SSL certificate via LetsEncrypt and added it to the server.
What I did:
openssl pkcs12 -export -out plex-certificate.pkfx -inkey /etc/letsencrypt/live/home.jhml.de/privkey.pem -in /etc/letsencrypt/live/home.jhml.de/cert.pem -certfile /etc/letsencrypt/live/home.jhml.de/fullchain.pem -passout pass:xxxxxxxx
mv plex-certificate.pkfx /ets/ssl/plex.pkfx
chown plex:plex /ets/ssl/plex.pkfx
I also setup the Variables like MovieFan suggested:
I restarted the plexmediaserver.service and tried again viy phone, but still no luck. Now the App on my phone is loading forever. If I try to set the streaming to “Play without transcoding” I get the same error as before: “There was an error while trying to play the video, Please check connection and try again”

I’m not positive on these things since I don’t know much about certificates, but I believe your certificate file is suppose to be a .p12 file, not pkfx. And the field for the encryption key is the file containing your key, not the actual key. The custom certificate domain should also just be the domain, no https and no port number.
Your making things harder on yourself by using these things. I would suggest first removing all this and just start with the certificate from Plex to make sure things are working. Then you can try adding your own certificate afterwards to be clear that it’s not a Plex issue, but to do with your certificate.
Based on your openssl command want to set the following for the 3 certificate fields as follows
Custom certificate location: /etc/letsencrypt/live/home.jhml.de/cert.pem
Custom certificate encruption key: /etc/letsencrypt/live/home.jhml.de/privkey.pem
Custom Certificate domain: home.jhml.de
And you will want to make sure that plex can read those files.

Over the last 2 Months I tried a lot off stuff. with certificates, without certificates… resetup my server, resetup the whole library…
It does not help.
Plex still refuses to play media files with a transcoder error, and I would like to fix this before I try to fix any other ‘possible’ problem.
Plex plays media files when I am connecteds to my home network. if I start to transcode files, it gives me an error.
I have a P2000 Quadro in my server which should habe enough power to transcode. drivers are installed and if i try a different software than plex, transcoding works just fine.
