How can i get my users to have custom profile picture’s?

I think that can only be done within the user’s Plex account preferences.

Each user can assign their own profile picture in their account settings:

  1. Open https://app.plex.tv/desktop#!/settings/account
  2. Click Edit on the Photo section
  3. Choose a photo or drag/drop one into the photo area
  4. Adjust the photo so it nicely fits the circle
  5. Save your changes

Is it possible to remove an assigned profile picture, and revert to the default?

Nope. You will have to find something, which you can use to replace the current avatar.
