If I change my user name using plex.tv/users/edit how does that affect Friends and Family shares that I have? Will it break them? How does that work since the shares were initially setup using my current user name?

Nope. Your username does not affect sharing.
How long does a user name change take to go into affect? I just tried to change mine and it shows the new user name in the edit user settings but if I log out I am unable to login with the new name. Only the old name will work.
The user name change goes into effect immediately.
Care must be taken to make sure the username you want isn’t already in use and you get the verification at the top of the page saying the change was successful. I edited my name and found a naming conflict. It confused me a bit at first.
A problem report to highlight an ‘already in use’ condition in red versus the current gray has already been submitted.
