I can direct play on MAC player by “remote access”.
But can’t direct play on the MiBox S / iPhone on local network(WiFi, need transcode).
And also can’t direct play on iPhone by “remote access”(need transcode))
Could someone tell me what the different for direct play?

A computer can typically play a larger number of video / audio types. Other devices including mobile phones have a limited number of video types that they can play directly so the Plex server will transcode videos or audio into a compatible type as needed.
Subtitles will also often require transcoding.
If you can post the actual media type (not just extension) we can let you know if it’s compatible with your iPhone or not.

The player is in the Plex player, so is it Plex player limitation?

Look up the device’s technical specifications for the supported audio and video formats. These vary from iPhone model somewhat for example. The information is available on Apple’s website.
On Mi Box did you set Video > Remote Streaming > Maximum? On iOS you would also similarly need to set Quality > Video Quality > Remote Streaming > Show All (at bottom) > Maximum. If those are indeed your settings then we will need the mediainfo of your files to further troubleshoot
