port 16621 is not a standard, fixed Plex port. That’s most likely UPnP and therefore transient. No need to add it.

On your modem/router, if it does not have UPnP/NAT-PMP,
Create a manual Port Forwarding rule to forward port 32400 to the IP address of your Syno also port 32400. Protocol is TCP. UDP is not required.
