I have about 25 or so smart collections for Actors. Made them by using a filter to get all movies for a given Actor, then saved it as a smart collection - i.e. “Movies - Actor Name”
Anyhow, a few months ago some of these have turned to 0, with no content in them.
Nothing has changed with the source files, path, or filename.
Does anyone know why this would be happening?
Most likely a side effect of this Plex is creating new/duplicate Tags for Directors and Actors that are already in my server - #6 by drzoidberg33

You probably need to edit the collection filter and save it (ideally no actual changes are needed but re-saving it should re-apply the correct actor).
I found when I changed the Poster for any movie in a collection they would fall out.
Then you probably have to newly select the actor/director in question

on my setup this doesn’t work.
if you just update the collection, it will update the cover images for the collection, but it will not re-populate it with the filtered content.
you essentially have to re-create the smart collection all over again.
Yes that is what i have experienced, you lose your custom poster. After that issue I now keep a Folder full of Custom Poster files just in case my Smart collections need a reset.
So if I am understanding this correctly, if you change the cover art for a movie in a smart collection it will then remove all content (i.e. movies) from that collection?

No, only if you recreate a Smart Collection or have a corrupt database that can not be repaired.
And just so I am clear on what is happening here:
you make a smart collection for (12) Tom Hanks movies in your library and save it.
you open Plex 2 days later and go to collections, and the Tom Hanks smart collection you made has no images on the cover, and instead of saying (12) it says (0)
When you click on the collection, there are no titles.
Then it is best to delete IMHO and recreate using the filters and custom metadata you have saved.
