Using USB external Hard drive docking station to stream media through Plex

Can someone give me a little advice or let me know if it is ok to stream some or most of my media library on my Plex server from a 6TB SATA External Hard Drive Docking Station and/or Enclosure?
I know that it will work, but I guess what I need to know is it going to be fast enough to stream a movie(s). I don’t have too many users yet, but plan to have more in the future and need to know if this would limit the amount of people should they all be streaming from that drive. I have 4 bays in the server, with 3 bays having 2TB drives for library content, which is only 6TB. The plan was to get the new 6TB drive in the server, but of course, I didn’t check compatibility and my hard drive controllers on the mother board only support up to 2TB Hard drives. Rather than wasting 4TB of drive space, I figured I would buy a External Hard drive enclosure or Hard drive docking station to make use of the full 6TB drive. I have a ton of back up USB drives, space already, so I really would want/need the 6TB to be used for media that will be streamed from USB 3.0, and not just for back up.
Anybody have any thoughts, suggestions or knowledge on if this is a viable option?
