I have Plex server running on Windows 10, if I map in media folders from a NAS how does Plex server handle this when a file is requested? I’m hoping that the server sends it directly from the NAS to whichever device is requesting it, or does it stream to my Windows computer first and send it back out again to the correct device? All my devices can handle 4K decoding locally so there should be no reason to go to the Windows PC first.

All playback goes through your server. That is what it is for, distributing files to the client.
