You need to enable the SMB server capability on the Shield.
See:
Plex Nvidia Shield Documentation
Specifically:
Media Storage Options for NVIDIA SHIELD
and
Accessing SHIELD Storage

Make sure you have the SMB v1 client enabled in Windows 10. It is disabled by default.
- Control Panel -> Programs and Features -> Turn Windows Features on or off
- Scroll down until you see
SMB 1.0/CIFS File Sharing Support. - Expand it and check the box for
SMB 1.0/CIFS Client. - Select OK to close the window.
- Windows will make the necessary additions to your system. The system should prompt you if a reboot is necessary.
It is not mentioned in the documentation, but the SMB server in the Shield only supports SMB 1.0 (the client does support SMB v3). Microsoft disabled SMB 1.0 in Windows 10. You have to enable it manually.
