I am no progammer and I have no programming knowledge but how difficult would it be to develop an application to run on either OS X or Windows to use AirPlay to point to the Plex server/home theater database and stream it to an AppleTV? Again I am sorry for the noob question, I'm just thinking out loud here :huh:

Not quite what you are talking about, but you can do this with an IOS device. If you are looking for something to stream directly from plex server, maybe a chromecast would work better for you?
I don't get it. download plex home theater, set it up for fullscreen mode on second screen and klick on Airplay Display: AppleTV - done. If your mac is too old, have a look at airparrot as mentioned by wahlmann.j
If you'd like to power down your mac like it's possible with an iOS device there is no chance, especially not with windows. Airplay uses OS X code that will not run on a windows pc, like you can't program iOS apps on a windows pc...

Yeah but I want to be able to use my Mac while my roommates may be in the other room watching a movie from my Mac.
Airplay direct from the server has been asked for in the past and because plex dont comment on features who knows if it is on the list of features being considered. Plexconnect is a community developed client that allows the ATV to connect to the PMS and present media, I would suggest you look at setting it up rather than wait for an airplay app as it is already here and working successfully.
(Personally i don't see how the request for an Airplay app has anything to do with the Plexconnect forum and as such should have been raised in the general forums)

If you have all your media on your Mac then install Plex Connect on the AppleTV3 or 2, or jailbreak your AppleTV2 and install Plex. Then there is no need for Airplay. If your media is on a server/nas there are options to airplay directly to your AppleTV like with my synology nas. Or does nobody have an iPhone/iPad/iPod of your roommates? Or even you? In my opinion an AppleTV makes just sense with other iOS devices... Then you could airplay to appleTV with one click und afterwords your roomates can control the AppleTV with the apple remote
