I know this is old... but there is a full SDK for integrating content into Windows Media Center, which would get Plex on the XBox as a WMC Extender. There's a couple "other" guys using this method: MyMovies and MediaBrowser to name a few of the ones I've tried. Nobody comes close to hitting the mark of Plex IMO. I'd gladly pay for a WMC Plex plugin.

WMC is a dead end, Microsoft are pulling the plug on that whole platform, it probably wont survive after Windows 8.
I think the odds of a Plex client might be better with the next generation Xbox, if Microsofts plan is a unified app ecosystem across PC's, phones, tablets then they may allow some degree of app development on the next Xbox.

Plex + Xbox360 sounds great but only if it’s possible outside of an xbox-live account. Xbox-live accounts retails for $59.99 a year ($35-$40 through Slickdeals), not a selling point if you don’t already use the service.

I would absolutely love this as well. Currently have a Mac Mini as a dedicated player, just for the gorgeous UI. A Plex App designed for Xbox would be incredible. #Plex4Lyf !
