So I have a folder “Power Book II” with “.plexignore” file and subfolder “Power Book II Ghost (Season 1) WEB-DL 1080p” with video files and also with “.plexignore”. Video files still persist in library.
Updating library files doesn’t help.

How can Ignore all files and subfolders of the folder where “.plexignore” file exists?
If it’s in a subfolder then the following will ignore everything in that subfolder.
# Ignore all subdirectories
*/
# Ignore all files
*.*
In your example you would only need one .plexignore file, preferably inside the folder where all the files are (so that you don’t accidentally ignore any new folders you put in there and wonder why they won’t show up).
So in that case just use do this inside the .plexignore file that live with the episode files:
# Ignore all files
*.*
