I’m rebuilding my Plex Media Server with some new hardware and I see a lot of users running PMS on Windows Server - so I’m wondering if there are specific advantages to running on Windows Server 2016 (with Desktop Experience) vs Windows 10 Pro?

No, there is no advantage of one over the other. In my case I have been running a server for years so it made sense to install it there.

It depends on your needs. Chances are if you don’t know why Server version is better than it won’t make any difference for you.
But in a nutshell if you are using ReFS, Storage Spaces, Virtualization, Shielded VMs, Active Director, etc then Server is the version you would want.
If your running in a “typical” home environment and have no special needs that require the server version then by all means just use Windows 10.
