Have not been able to find anything yet that explains it
It appears that Plex Media Server.exe is able to run this C:\Program Files (x86)\Plex\Plex Media Server\PlexScriptHost.exe
but not C:\Program Files (x86)\Plex\Plex Media Server\Plex Tuner Service.exe
or this C:\Program Files (x86)\Plex\Plex Media Server\Plex DLNA Server.exe
Could you see if they got quarantened by antivirus / defender
you have this running MsMpEng.exe and also SmartScreen.exe
I don’t know, I disabled smart screen and real-time protection and anything else security related. Restarted Plex Media Server as admin and still had the problem. I guess my next step is to backup my database and build a new server and see what happens. Very odd.
I scanned with Malwarebytes to see if there was some sort of infection and it found nothing, which is expected since I don’t do anything with this box except for run Plex.
It is strange that this can be run by Plex Media Server C:\Program Files (x86)\Plex\Plex Media Server\PlexScriptHost.exe
but not these two
C:\Program Files (x86)\Plex\Plex Media Server\Plex Tuner Service.exe
C:\Program Files (x86)\Plex\Plex Media Server\Plex DLNA Server.exe
What shows in Windows Defender Firewall Advanced Settings under “Monitoring → Firewall” view for these - Should have an “Allow” for each
Does the “Virus and threat protection” view (Run/Virus and threat protection ) show anything? eg in Protection History
You can try a repair of the Installation of Plex Media Server through Add or Remove Programs - or uninstall and re-install - installing latest beta. and see if we still get the Access Denied error logged in the Plex Media Server log

I have done an uninstall and re-install recently, but it didn’t help. That’s why I said I might just have to build a new server and backup and restore my PMS database. I may try running these manually and see what I get:
