I recently got an Nvidia Shield Pro (about a month now). When I first set it up, Plex was an option in the settings for “Play Next” on the Home screen. And over the last week or two, that section stopped updating, and Plex is no longer an option in the Home settings menu. I really liked that feature.
Same issue here. I think it was the last update that broke it. Both the ‘Play Next’ and ‘Recommended’ home screen channels stopped updating on my Shield Pro. After reinstalling plex (thinking it had become corrupted somehow), it has not come back. No Play Next, and no Recommended. Going into the ‘Customize channels’ menu in Android TV doesn’t show Plex as an option anymore.
Same issue here, also uninstalled/reinstalled app thinking it would help and since doing so have now lost Plex from the customise channels and play next sections for Home screen. Hopfully they fix soon 
If you have not, can you check for the update to 9.21.0.1123. Play Next is not something i normally use but Plex was showing up for me in the Play Next settings and the row appeared on home screen after I enabled it.
Mine automatically updated to 9.21.0.1123 yesterday. Still has the same issue. I’ve just tried reinstalling again but still no home screen channels. The option to add them is also still not there unfortunately.

Same for me. Mine also auto-updated, and the behavior didn’t improve.
Just to be clear for other folks who may see this topic. Also I realized i am on an older original shield so that may be the difference for me
You do not see this to enable correct? or is it there and you have it enabled but it does not show up on home screen?

Yep, same here. No longer an option. Also does not appear in the ‘Home screen channels’ section of the Customize channels menu (for Recommended etc.).
I have the same issue on regular Android TV running 9.21.1.1169.

Same here. Disappeared. Casting options also disappeared can’t cast from my phone.
Not sure what you mean by casting disappeared, since Plex doesn’t really participate in the ChromeCast/Android TV casting feature. Regular casting still works fine for me.
Once you uninstall and reinstall the Plex Android TV app you can no longer remote control the Android TV app using the other Plex apps. Casting using Chromecast built-in still works fine.
